Magnesol XL concentration (0.5, 1, 3 and 5%), heating temperature (25, 50, 70 and 90 ˚C) and time (5, 10, 15 and 20 mnt) during purification to the color properties (Lightness L*, redness a* and yellowness b*) of Sardinella lemuru oil were evaluated. Purification using Magnesol XL in any condition effectively increase the L* and a* value but reduced the b* value of the lemuru oil. Highest L* value (96.57) was achieved at the treatment temperature 90 ˚C, 5 % level of Magnesol XL concentration and 5 minutes process. Lowest a* value (more green color) was obtained at treatment 70 ˚C temperature, 5% level of concentration and 15 minutes, then lowest b* value was obtained at treatment 90 ˚C temperature, 5 % concentration and 5 minutes process. All the refined lemuru oil’s result had a hue angle higher than 90˚ representing the light greenish-yellow color.
Proteases were extracted from starfruit at maturity Index 2 (unripe, light green) and Index 7 (very ripe, orange) and partially purified using acetone and 40% ammonium sulfate precipitations. Higher yield and proteolytic activity were observed for proteases purified using acetone than 40% ammonium sulfate. As for maturity index, yield and protein concentration of proteases from Index 2 were higher than those from Index 7. SDS-PAGE result showed intense bands for acetone proteases while a distinct band at 50 kDa was observed in all the proteases. Enzyme activity decreased during the seven days storage at 4°C with minimum relative activity of 70% achieved for acetone proteases at day seven. This study suggested that acetone precipitation is more effective method for purifying starfruit protease based on the yield and proteolytic activity compared to using 40% ammonium sulphate precipitation. In order to obtain higher protein concentration and proteolytic activity, starfruit at the unripe stage, Index 2 is a better raw material than Index 7 to be used for protease production.
Blastocystis hominis has been regarded as an enigmatic parasite as many aspects of its basic biology remain uncertain. Many reproductive processes have been suggested for the organism; however, to date, only the binary fission has been proven. Plasmotomy is one of the modes of reproduction previously suggested to be seen in in vitro cultures. The present study provides trichrome and acridine orange staining evidence for the existence of nucleic acid suggestive of division of nucleus into multinucleate forms with the respective cytoplasm dividing giving rise to two or three progeny B. hominis. Transmission electron micrographs further confirmed that these daughter cells had respective surrounding surface coat, mitochondria, and vacuoles.
Recycling of alternative water sources particularly greywater and recovery of energy from wastewater are gaining momentum due to clean water scarcity and energy crisis. In this study, the photocatalytic fuel cell (PFC) employing ZnO/Zn photoanode and CuO/Cu photocathode was successfully designed for effective greywater recycling as well as energy recovery. The photoelectrodes were analyzed using X-ray diffraction (XRD), field-emission scanning electron microscopy (FESEM), energy dispersive X-ray (EDX) and fourier transform infrared (FTIR) spectroscopy. The PFC performance in terms of electricity generation and parallel methyl green (MG) degradation were evaluated under operating parameters such as electrolyte type, initial MG concentration and solution pH. The results showed that the addition of Na2SO4 electrolyte, MG concentration of 40 mg L-1 and solution pH of 5.2 improved the short circuit current density (Jsc) and power density (Pmax) in the as-constructed PFC. Such a system also afforded highest MG and chemical oxygen demand (COD) removal efficiencies after 4 h of irradiation. The photoanodes used in this study demonstrated great recyclability after four repetition tests. The COD removal was reduced to some extents when the PFC treatment was tested in the real greywater under optimal conditions. Various greywater quality parameters including ammoniacal nitrogen (NH3-N), turbidity, pH and biochemical oxygen demand (BOD5) were also monitored. The phytotoxicity experiments via Vigna radiate seeds indicated a reduction in the phytotoxicity.
