    Physical training is associated with oxidative stress and improvement in blood antioxidant status. In this study, we investigated the effects of training on plasma malondialdehyde (MDA) and 4-hydroxynonenal (4-HNE) as markers of lipid peroxidation, superoxide dismutase (SOD) activity, and ascorbic acid (AA) after a single bout of acute exercise. Twelve healthy, untrained young adult men were recruited for 5 weeks of aerobic training period. They were subjected to a rope-skipping exercise for 20 minutes at the intensity of 65-80% of heart rate reserve, thrice weekly. They also had to perform a single, acute bout of the same exercise protocol prior to and after training period. Venous blood samples were collected at resting condition (BL), immediately (0 h) and 24 hours (24 h) post acute exercise on both single bout sessions. Results showed that the pattern changes of oxidative stress response are quite similar on both acute sessions. The acute bouts of rope-skipping is associated with a significant increased (p = 0.001) in lipid peroxidation markers immediately after cessation of exercise, with a concomitant increases in antioxidant levels, albeit higher when compared to pre-training values (p = 0.001). These changes were followed by a significant decreased (p = 0.001) in all parameters, toward resting values, 24 hours thereafter. The training program seemed to induce a significant increase in MDA and 4-1-INE but also enhanced the antioxidant defense system namely SOD and AA among the untrained subjects.

    Abundant natural products with medicinal properties have been used as food and traditional medicine for diabetes mellitus all over the world. Psidium guajava fruit from the family of Myrtaceae has gained attention for its antioxidant potential. This study was conducted to determine the effects of P. guajava fruit peel aqueous extract on oxidative stress of pancreas in streptozotocin-induced (45 mg/kg) diabetic rats. Diabetic rats were administered with 400 mg/kg of aqueous extract of P. guajava fruit peel daily for 28 days duration. The results showed that diabetic rats supplemented with P. guajava extract did not cause significant difference in blood glucose level (p>0.05) as compared with diabetic rats alone. For oxidative stress evaluation, malondialdehyde (MDA) and protein carbonyl level were significantly lower and the activity of superoxide dismutase (SOD) and glutathione (GSH) level were significantly higher (p<0.05) in P. guajava supplemented rats compared with non-supplemented diabetic rats. However, histological observation showed that supplementation of P. guajava extract did not give protective effects towards alterations in pancreas histology in diabetic rats. The findings suggested that aqueous extract of P. guajava fruit peel supplementation has the ability to reduce oxidative stress in pancreas of diabetic rats and may play a role in reducing the development of diabetic complications.

    Background: Sudden death (SD) is defined as an unexpected natural death within an hour onset of symptoms or unwitnessed death that occurs within 24 hours, which accounts for most cardiovascular deaths in Malaysia. This study aimed to evaluate the extent of histopathological changes in the heart and to study the demographic trend in SD cases in Kuala Lumpur, Malaysia. Methods: Specimens from all SD cases were received from 2017 to 2018 by the Forensic Unit, Hospital Canselor Tuanku Mukhriz (HCTM) were studied. The specimens were the left ventricular myocardium and left anterior descending artery (LAD), which were fixed in 10% formalin with haematoxylin and eosin staining. The tissues were graded histologically based on changes such as arterial occlusion, myocardial infarction, and/or thrombus formation. Results: Out of 545 medicolegal deaths, only 25 cases (4.6%) had samples available for analysis. Among these 25 patients, 24 (96%) were male patients and only one (4%) was a female patient. The available samples were from patients aged between 30 to 79 years old. In terms of ethinicity, Malays (40%) were the most numerous, followed by patients of Chinese and Indian ethnicities.The majority of the SD cases had a body mass index (BMI) that ranged between underweight (56%) and obese (40%). Besides, there were 10 (40%) cases of coronary artery with atheroma and 15 (60%) cases of myocardial infarction. The most common presentation of atherosclerosis was grade III and IV, and acute MI was the most common presentation at death, followed by healed infarcts and old infarcts. Discussion and Conclusion: Our findings reflect worsening risk factor levels in cardiovascular diseases, compounded by demographic trends. Further studies on biomarkers specific for cardiac diseases are warranted to understand imminent sudden cardiac death.

    Background: The main mechanism of fenitrothion (FNT) toxicity is by inhibiting the acetylcholinesterase enzyme, though studies have shown that FNT might also develop other toxicological manifestations, including oxidative stress. However, the kidney functions as well as oxidative stress status after repeated exposure to FNT is not well documented. Objective: The present study was conducted to evaluate the kidney functions and oxidative stress status after exposure to low doses of FNT. Materials and Methods: 24 male Sprague-Dawley rats were divided randomly into three groups (n = 8/group): control, which received 1 mg/ml corn oil; FNT 10, which received 10 mg/kg (BW) FNT; and FNT20 which received 20 mg/kg (BW) FNT. The corn oil and FNT were fed orally for 28 consecutive days. At the end of the study, the blood was taken, and the kidney was obtained for biochemistry evaluation and histological observation. Results: The AChE activity was significantly inhibited in the FNT20 group (p≤0.05) compared to the control group and FNT10 and the rats exhibited the signs and symptoms of toxicity such as lacrimation, piloerection, hypoactivity, and tremor. Plasma creatinine and BUN levels showed a significant increase (p≤0.05) in FNT treated groups, but the superoxide dismutase and glutathione level were significantly reduced (p≤0.05). The malondialdehyde and protein carbonyl level were elevated significantly (p≤0.05) in FNT treated groups. Histopathological observation revealed morphological changes, including atrophy of the glomerulus and presence of non-amyloid substances in FNT treated groups. Conclusion: Even at the dose that did not inhibit the AChE activity, FNT was found to reduce the renal function and induce oxidative damage on the kidney of male Sprague Dawley rats.

    The fruit of Canarium odontophyllum Miq. is a traditional delicacy in Borneo for its anti-aging benefit. This study evaluated the protective effect of C. odontophyllum leaf aqueous extract on damaged liver in streptozotocin-induced diabetic rats. A total of 30 male Spraque-Dawley rats (150-250g) were randomly divided into three groups: control group, diabetic without treatment and diabetic treated with 300 mg/kg aqueous extract of C. odontophyllum for 28 consecutive days. The diabetic condition was induced by intraperitoneal injection of streptozotocin at 65 mg/kg body weight. At the end of study period, blood was collected to assess the biochemical changes and the oxidative stress markers whereas the liver section was examined for morphological changes.     The relationship between anaemia and cognitive function was evaluated among 35 Chinese elderly (24 men and 11 women) aged 60 to 85 years (mean age 70.1 ± 6.7 years) from five old folks homes in Klang Valley. They were interviewed to obtain information on social and health status, habitual dietary intake and cognitive function. Hodkinson's Abbreviated Mental Test was used to measure the cognitive function. Haematological indices which included Full Blood Count (FBC), serum iron, serum ferritin, Total Iron Binding Capacity (TIBC), serum folate and serum cobalamine (vitamin B12) were measured using an automated analyzer. Anthropometric measurements and clinical signs of anaemia were also examined. The findings indicated that the prevalence of anaemia as assessed using haemoglobin alone was 22.9%, while iron deficiency anaemia based on low serum iron, microcytic and hypochromic criterion was detected among 5.7% of the sample. Subclinical folate and vitamin B12 deficiencies were diagnosed among 34.3% and 8.6% of the subjects. However, there was no occurrence of megaloblastic anaemia.     The protective effect of the ethanol extract of the rhizome of Zingiber officinale Roscoe on acute hepatotoxicity induced by paracetamol (1000 mg/kg) was studied in plasma and hepatic tissue samples obtained from male Sprague-Dawley rats. The ethanol extract was given in oral doses of 200 mg/kg and 300 mg/kg to the rats at 0, 4 and 8 hrs after paracetamol was given orally. The plasma and liver of the rats were subjected to biochemical analysis 24 hrs after hepatotoxicity was induced to determine the levels of superoxide dismutase(SOD), malonaldehyde (MDA) and aspartate transaminase (AST). The results were compared to the rats which were given the antidote N-acetylcysteine (NAC) (500 mg/kg) at 0, 4 and 8 hrs after the paracetamol dose. The results showed that at 200 mg/kg the extract reduced the plasma levels of SOD significantly (p < 0.05) while at a higher dose of 300 mg/kg it reduced plasma SOD, hepatic MDA, serum AST and increased the levels of plasma proteins significantly (p < 0.05). In conclusion, the ethanol extract of Z. officinale showed protective effect against paracetamol induced hepatotoxicity at both dose levels and the protective effect was better at the higher dose.

    Exposure to organophosphate pesticide including fenitrothion (FNT) has led to many adverse effects on human health.
    However, a potent antioxidant like palm oil tocotrienol-rich fraction (TRF) can reduce oxidative damage in various
    pathological conditions, could also reduce the adverse effects by FNT. The aim of this study was to evaluate the effect
    of TRF on oxidative liver damage in FNT induce hepatotoxicity in experimental rats. A total of 40 male Sprague-Dawley
    rats were randomly divided into four groups of 10, namely control, TRF, FNT and TRF+FNT group. TRF (200 mg/kg
    body weight) and FNT (20 m/kg body weight) were administered through oral gavage for 28 days. Corn oil which
    served as vehicle was given orally to the control group. At the end of the study period, liver and blood was taken for
    oxidative damage and biochemical evaluation and histological observation. TRF supplementation prevents oxidative
    liver damage by reducing the hepatic malondialdehyde (MDA) and protein carbonyl (PCO) level significantly. Besides,
    TRF also restored the endogenous antioxidants particularly reduced glutathione (GSH), glutathione peroxidase (GPx)
    and ferric reducing/antioxidant power (FRAP). TRF also prevent liver damage by reducing the liver enzymes, alanine
    aminotransferase (ALT) and aspartate aminotransferase (AST). The attenuation of liver damage by TRF was also showed
    histologically. In conclusion, TRF supplementation showed a potential in preventing oxidative liver damage in FNTtreated
    rats by reducing the oxidative damage and improving the antioxidant status.

    Penghasilan radikal bebas oleh nikotin dikaitkan dengan kerosakan sistem pembiakan lelaki terutamanya sperma dan testis. Penggunaan rawatan yang berasaskan herba seperti Hibiscus sabdariffa Linn. (HSE) kian meningkat disebabkan kandungan antioksida semula jadi yang tinggi. Oleh itu, kajian ini dijalankan untuk mengkaji kesan ekstrak akueus HSE terhadap kualiti sperma dan tekanan oksidatif testis tikus yang diadministrasi nikotin. Sejumlah 21 ekor tikus jantan Sprague-Dawley dibahagikan secara rawak kepada tiga kumpulan iaitu kumpulan kawalan, nikotin dan nikotin+HSE. Nikotin disuntik secara intraperitoneum pada dos 0.6 mg/kg berat badan manakala HSE diberikan pada dos 100 mg/kg berat badan secara paksaan oral sebelum administrasi nikotin pada setiap hari selama 21 hari berturut-turut. Hasil kajian menunjukkan bilangan, motiliti dan viabiliti sperma lebih tinggi secara signifikan (p<0.05) manakala peratus ketaknormalan morfologi sperma lebih rendah secara signifikan (p<0.05) bagi pada kumpulan nikotin+HSE berbanding kumpulan nikotin. Sementara itu berlakunya penurunan aras malondialdehid (MDA) dan peningkatan aras glutation terturun (GSH) secara signifikan (p<0.05) bagi kumpulan nikotin+HSE berbanding kumpulan nikotin. Pemerhatian histologi mendapati HSE berpotensi melindungi morfologi testis tikus aruhan nikotin. Kesimpulannya, kajian ini menunjukkan bahawa pemberian suplemen ekstrak HSE berpotensi mencegah kerosakan sperma dan testis akibat administrasi nikotin.

    UKMR-1, a local variant of mutant Roselle strain (Hibiscus sabdariffa) is enriched with free radical scavenging polyphenols
    such as anthocyanin, vitamin C and hydroxycitric acid. However, pharmacological actions of UKMR-1 are not fully known.
    This study was conducted to determine whether supplementation of aqueous UKMR-1 calyx extract was able to protect
    against nicotine-induced cardiac injury in rats. In this experimental study, healthy male albino rats were randomly
    allotted into three groups (n=7 per group): control, nicotine and UKMR-1+Nicotine groups. Nicotine (0.6 mg/kg, i.p.)
    was administered to both nicotine and UKMR-1+Nicotine groups for 28 consecutive days. UKMR-1+Nicotine group also
    received 100 mg/kg UKMR-1 extract orally via gavage 30 min prior to nicotine injection, daily. UKMR-1+Nicotine group
    had significantly (p<0.05) higher lactate dehydrogenase (LDH) activity, as well as lower malondialdehyde content in
    heart tissue homogenate than nicotine group, suggesting its cardio protective activity by inhibition of lipid peroxidation.

    Nicotine has been identified as one of the causal factor for oxidative stress, hypertension and hyperlipidemia. Roselle has been widely studied for its potential as an antioxidant, antihyperlipidemic and antihypertensive. However, no studies have been done to investigate if roselle could diminish the oxidative stress caused by nicotine which could further lead to cardiac damages. Thus, this study was aimed to investigate the effect of roselle extract (HSE) on blood pressure, serum lipid profile, oxidative stress marker levels and histological changes to the heart in nicotine-treated rats. A total of 21 Sprague-Dawley rats were randomly divided into 3 groups (n=7 per group): Control group received normal saline (0.5 mL/day, i.p); nicotine group received 0.6 mg/kg/BW nicotine (i.p); and treated group received 100 mg/kg/b.w HSE through oral force feeding followed with 0.6 mg/kg/b.w nicotine (i.p) for 21 consecutive days. The results showed that HSE significantly (p>0.05) reduced the heart rate but no effect to the blood pressure. For lipid profile study, HSE increased the high-density lipoprotein concentration significantly (p<0.05) in rats given with nicotine, without any significant changes in total cholesterol, triglyceride and low-density lipoprotein (LDL) concentration. Besides, HSE treatment was also found to reverse malondialdehyde (MDA) level, superoxide dimustase (SOD) enzyme activity and protein concentration significantly (p<0.05) in nicotine-treated rats. In summary, these results indicated that HSE is an effective antioxidant against oxidative damage in heart caused by nicotine, but not as antihyperlipidemic and antihypertensive agent in this rat model.

    This study was conducted to evaluate the oxidative damage in diabetic mellitus induced rats. The evaluation of DNA damage was carried out by the Alkaline Comet Assay using peripheral lymphocyte cells taken from streptozotocin-induced diabetic rats (50 mg/kg) and control rats. The levels of malondealdehyde (MDA), 4-hydroxynonenal (4-HNE), fasting blood glucose (FBG) and HbA1c were also measured. All the induced diabetic rats were hyperglycemic until the end of the study with significantly higher levels of FBG and HbA1c as compared to the control rats. The results showed the percentage of tail DNA and tail moment values were also significantly higher in the diabetic induced rats. The same observations were made on the levels of plasma MDA and 4-HNE. In conclusion, this study indicated that hyperglycemic condition in diabetic induced rats could generate oxidative DNA damage.

    Disturbances in immune system contribute to chronic infection among diabetic patients. Hibiscus sabdariffa L. (roselle) fruit extract has been scientifically proven to possess antioxidant, antidiabetic and antiinflammatory properties. The aim of this study was to investigate the effects of H. sabdariffa fruit extract against oxidative stress parameter and T lymphocyte population in spleen of streptozotocin (STZ) induced diabetic rats. Male Sprague-Dawley rats were injected with 45 mg/kg STZ to induce diabetic condition and further treated with 100 mg/kg H. sabdariffa fruit aquoeus extract daily for 28 days. Spleen was harvested to determine the oxidative stress indicators and quantification of T lymphocytes. The results showed a significant decreased in the number of spleen cells and spleen weight in the diabetic rats compared with control rats. However, there were no significant changes in the level of malondialdehyde (MDA), protein carbonyl and superoxide dismutase (SOD) activity the percentage of spleen CD3+ CD4+ and CD3+ CD8+ T lymphocytes amongst groups of study. In addition, histology observation showed no pathological alteration in spleen histology of diabetic rats. The findings suggested that aqueous extract of H. Sabdariffa fruit supplementation has no effect on the oxidative stress and the percentage of CD3+ CD4+ and CD3+ CD8+ T lymphocytes in spleen of diabetic rats.

    Fenitrothion (FNT) usage has received much attention for its potential to promote free radicals generation and interfere with antioxidant defense system. The aim of the present study was to investigate the effect of palm oil tocotrienol rich fraction (TRF) supplementation on oxidative stress and histological changes in rat brain induced by FNT. A total of 32 male Sprague Dawley rats divided into four groups: control group which received corn oil; TRF group was received palm oil TRF (200 mg/kg bw); FNT group administered with FNT (20 mg/kg bw) and TRF+FNT group pretreated with palm oil TRF (200 mg/kg bw) 30 min prior to administration of FNT (20 mg/kg bw). FNT and TRF were dissolved in corn oil and all supplementations were given by oral gavage once daily for 28 days. After four weeks of supplementation, TRF+FNT rats had significantly lower malondialdehyde (MDA) content and superoxide dismutase (SOD) activity but higher reduced glutathione (GSH) level and total protein level compared to FNT rats (p<0.05). However, protein carbonyl (PC) level was insignificantly lower for TRF+FNT group compared to FNT group. In conclusion, this study suggested that palm oil TRF was effective in preventing brain damage in rats.

    In this study, the effects of palm vitamin E (PV) supplementation on glycemic control and lipid profile in diabetic-induce Sprague-Dawley rats have been evaluated. Diabetes in the rats was induced by a single intravenous streptozotocin (50 mg/kg body weight). The diabetic rats were divided into two groups; supplemented with 200 mg/kg body weight/day of PV and non-supplemented with PV (No PV group). Non-diabetic rats (NDM) formed the control group and only received saline injection.
