    Gel- based proteomics is one of the most versatile methods for fractionating protein complexes. Among these methods, two dimensional- polyacrylamide gel electrophoresis (2-DE) represents a mainstay orthogonal approach, which is popularly used to simultaneously fractionate, identify, and quantify proteins when coupled with mass spectrometric identification or other immunological tests. Although 2-DE was first introduced more than three decades ago, several challenges and limitations to its utility still exist. This review discusses the principles of 2-DE as well as both recent methodological advances and new applications.

    Vulnerable communities in Malaysia have been facing issues of accessibility and availability for pediatric cardiac services for years due to long waiting times, high costs and a lack of pediatric cardiothoracic surgeons. To ease this situation, the government has allocated a certain amount of funds, introduced through the Pediatric Cardiothoracic Program (PCP), in which the patients are transported to the Narayana Health Institute of Cardiac Science (NH) in India to receive a heart operation following an eligibility check at MediAssist4U Sdn Bhd in Selangor, Malaysia, a facility appointed by the NH. This study aimed to determine the demographic incidence of congenital heart diseases of patients in this program and to evaluate the outcome of the program in association with post-operative mortality rate and the beneficial factors of the program. In this study, 241 patients who participated in this program from August 2008 to September 2012 were reviewed. Fisher's exact tests were applied to calculate p-values of categorical data. Out of 241, 11 patients were rejected because of their poor health condition for flight transportation to India, leaving 230 patients for analysis. The majority of patients were 1 to 4 years of age (57.8%), Malays (61.7%), from families of monthly household income less than RM 1,500 (86.5%) and with primary school-educated parents (86.5%). Patients could apply from any government hospital in Malaysia, but 34.8% of the patients were from the state of Johor. The region (Peninsular Malaysia and East Malaysia) of patients seeking pediatric cardiac services was significantly associated with race (p<0.001), low household income (p<0.001) and low education background of parents (p=0.004). The associations between the age group and diagnosis group (p=0.010) and between duration of hospitalization and outcome of medical management (p=0.013) were significant. Post-operative mortality rate was 1.7% (95% confidence interval, 0.5-4.4). In conclusion, the patients and the government were considered to have benefited from the PCP.

    A 240-nucleotide sequence of the capsid/premembrane gene region of 23 Japanese encephalitis virus (JEV) strains isolated in Tokyo and Oita, Japan was determined and phylogenetic analyses were performed. All the strains clustered into two distinct genotypes (III and I). All strains isolated before 1991 belonged to genotype III, while those isolated after 1994 belonged to genotype I. In addition, the strains of the genotype I isolated in Japan showed a close genetic relationship with those from Korea and Malaysia.

    Inverse vulcanized polysulfides (IVP) are promising sulfur-enriched copolymers with unconventional properties irresistible for diverse applications like Hg2+ remediation. Nevertheless, due to their inherent hydrophobic nature, these copolymers still offer low Hg2+ uptake capacity. Herein, we reported the synthesis of IVP by reacting molten sulfur with 4-vinyl benzyl chloride, followed by their functionalization using N-methyl D-glucamine (NMDG) to increase the hydration of the developed IVP. The chemical composition and structure of the functionalized IVP were proposed based on FTIR and XPS analysis. The functionalized IVP demonstrated a high mercury adsorption capacity of 608 mg/g (compared to <26 mg/g for common IVP) because of rich sulfur and hydrophilic regions. NMDG functionalized IVP removed 100 % Hg2+ from a low feed concentration (10-50 mg/l). A predictive machine learning model was also developed to predict the amount of mercury removed (%) using GPR, ANN, Decision Tree, and SVM algorithms. Hyperparameter and loss function optimization was also carried out to reduce the prediction error. The optimized GPR algorithm demonstrated high R2 (0.99 (training) and 0.98 (unseen)) and low RMSE (2.74 (training) and 2.53 (unseen)) values indicating its goodness in predicting the amount of mercury removed. The produced functionalized IVP can be regenerated and reused with constant Hg2+ uptake capacity. Sulfur is the waste of the petrochemical industry and is abundantly available, making the functionalized IVP a sustainable and cheap adsorbent that can be produced for high-volume Hg2+ remediation. ENVIRONMENTAL IMPLICATION: This research effectively addresses the removal of the global top-priority neurotoxic pollutant mercury, which is toxic even at low concentrations. We attempted to remove the Hg2+ utilizing an inexpensive adsorbent developed by NMDG functionalized copolymer of molten sulfur and VBC. A predictive machine learning model was also formulated to predict the amount of mercury removal from wastewater with only a 0.05 % error which shows the goodness of the developed model. This work is critical in utilizing this low-cost adsorbent and demonstrates its potential for large-scale industrial application.

    AIM: To evaluate the dental pulp response to a novel mineral trioxide aggregate containing phosphorylated pullulan (MTAPPL) in rats after direct pulp capping.

    METHODS: Ninety-six cavities were prepared in the maxillary first molars of 56 male Wistar rats. The dental pulps were intentionally exposed and randomly divided into four groups according to the application of pulp capping materials: MTAPPL; phosphorylated pullulan (PPL); a conventional MTA (Nex-Cem MTA, NCMTA; positive control); and Super-Bond (SB; negative control). All cavities were restored with SB and observed for pulpal responses at 1-, 3-, 7- and 28-day intervals using a histological scoring system. Statistical analysis was performed using Kruskal-Wallis and Mann-Whitney U-test with Bonferroni's correction, and the level of significance was set at 0.05. DMP1 and CD34 antigen were used to evaluate odontoblast differentiation and pulpal vascularization, respectively.

    RESULTS: On day 1, mild inflammatory cells were present in MTAPPL and NCMTA groups; fewer inflammatory cells were present in the PPL, whereas SB was associated with a mild-to-moderate inflammatory response.
