    The bioavailability of a generic diclofenac sodium sustained release tablet preparation (Zolterol, SR) was compared with the innovator product, Voltaren, SR. Twelve healthy adult male volunteers participated in the study, which was conducted according to a randomized, two-way crossover design with a wash out period of one week. The bioavailability of diclofenac was compared using the parameters area under the plasma concentration-time curve (AUC(0-infinity)), peak plasma concentration (Cmax) and time to reach peak plasma concentration (Tmax). No statistically significant difference was observed for both logarithmically transformed AUC(0-infinity), Cmax values and Tmax value of the two preparations.

    In this research, the Cu-based metal-organic framework (MOF-199) was fabricated and coated on the stainless steel mesh as substrates through sol-gel procedure. Then the coated substrates were placed in a small column known as solid-phase extraction cartridge. The SPE based coated stainless steel mesh coupled with high-performance liquid chromatography-UV detector (HPLC-UV) was used for the fast extraction, and quantification of non-steroidal anti-inflammatory drugs (NSAIDs) from human plasma and water samples. To find optimum extraction conditions, the impacts of effective parameters on analytical performance like sample pH, sample volume, type, and volume of desorption solvent were optimized. At the optimized conditions, calibration graphs of analytes were linear in the concentration range of 0.03-300 ng mL-1 for water samples, and 0.1-200 ng mL-1 for plasma samples. The correlation coefficients were in the range of 0.9938 to 0.9989. Also, the limits of detection (LODs) were from 0.01 to 0.02 ng mL-1 for water samples and 0.03 to 0.1 ng mL-1 for plasma samples. The cartridge repeatability was studied at different values, and the relative standard deviations (RSDs%) were achieved between 3.5 and 5.1%. Consequently, this procedure was successfully used in the extraction and detection of NSAIDs in real water and plasma samples with relative recoveries ranged from 93.6 to 99.6%.

    A xanthan gum matrix controlled release tablet formulation containing diclofenac sodium was evaluated in vitro and was found to release the drug at a uniform rate. The gastrointestinal transit behaviour of the formulation as determined by gamma scintigraphy, using healthy male volunteers under fasted and fed conditions, indicated that gastric emptying was delayed with food intake. In contrast, the small intestinal transit remained practically unchanged under both food statuses. Therefore, the delay in caecal arrival observed in the fed state can be attributed to the delay in gastric emptying. Rate of diclofenac sodium absorption was generally higher in the fed state compared to the fasted state, however the total amount absorbed under both food statuses remained practically the same. The rate of in vivo dissolution of the drug in the fed state was faster compared to that in the fasted state. Thus, at the time of caecal arrival, in vivo dissolution was complete in the fed state, unlike in the fasted state, where almost 60% of the drug was delivered to the colon.

    A simple and validated high-performance liquid chromatography (HPLC) method with UV detection has been used to determine the content of andrographolide (AP) and 14-deoxy-11,12-didehydroandrographolide (DIAP) in rat plasma after oral dose of methanol extract (1 g/kg body weight) of Andrographis paniculata leaf. An increase in plasma concentration of AP and DIAP was observed from 30 min to 3 h after oral administration of the extract. The maximum plasma concentrations of AP and DIAP were 1.42+/-0.09 microg/ml and 1.31+/-0.04 microg/ml, respectively. Fourteen days oral treatment of rats with the methanol extract (1 g/kg body weight) followed by CCl(4) administration preserved catalase (CAT), and superoxide dismutase (SOD) activities in erythrocytes, whereas plasma lipid peroxidation, alanine transaminase (ALT) and aspartate transaminase (AST) activities were restored to values comparable with control values. Treatment of rats with CCl(4) did not showed significant alteration (p>0.05) in plasma total antioxidant status (TAS) as compare to values of control group.
