    The methanol extract of the dried ripe fruits of Alpinia rafflesiana was investigated for its DPPH free radical scavenger constituents. 2',3',4',6'-Tetrahydroxychalcone (7), which has never been isolated from natural sources was found to be most active as a DPPH free radical scavenger with the IC50 value of 55 microM. Other known compounds isolated from this species include 5,6-dehydrokawain (1), flavokawin B (2). 1,7-diphenyl-5-hydroxy-6-hepten-3-one (3), (-)-pinocembrin (4), cardamonin (5) and (-)-pinostrobin (6). The DPPH free radical scavenger compounds were detected using TLC autographic analysis. The percentage inhibition of DPPH free radical scavenging activity was measured on isolates (5-7) using colorimetric analysis.

    Antioxidant capacities of ethylacetate, butanol, and water fractions of peel, pulp, and seeds of Canarium odontophyllum Miq. (CO) were determined using various in vitro antioxidant models. Ethylacetate fraction of peel (EAFPE) exhibited the highest total phenolic (TPC), total flavonoid content (TFC), and antioxidant activities compared to pulp, seeds, and other solvent fractions. Antioxidant capacities were assayed by total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical activity, ferric reducing antioxidant power (FRAP), and hemoglobin oxidation assay. Total phenolic content of ethylacetate fractions was positively correlated with the antioxidant activity. This is the first report on the antioxidant activities from CO fruit fractions. Thus, EAFPE can be used potentially as a readily accessible source of natural antioxidants and as a possible pharmaceutical supplement.

    Plant tissue or cell culture keeps a significant role in micro-propagation in the plant production industry. Combination of 6-Benzylaminopurine (BAP) and other plant growth regulators like 1-Naphthaleneacetic acid (NAA) or Indole-3-acetic acid (IAA) or indole-3-butyric acid (IBA) was used in the most of the research in tissue culture. The study was carried out to investigate the optimization of the concentration of IBA and BAP combination (0, 0.25, 0.50, 1.0, 1.50, 2.0, 2.5, 3.0 and 3.5 mg/l) for the root, callus and leaf proliferation from the leaf cutting slice. The highest number (6.75) of root proliferation was observed in the concentration of 2.0 mg/l IBA+0.25 mg/l BAP combination. The callus initiation was found in the concentration of IBA 1.0-3.5 mg/l+BAP 1.0-2.0 mg/l. However, the highest callus weight was observed at the concentration of IBA 1.5 mg/l+BAP 1.0 mg/l combination than other combination of concentrations. Positively leaf initiation and formation was better in the concentration of IBA 1-3.5 mg/l+BAP 1.0-2.0 mg/l combination. In addition, the 2,2-diphenyl-2-picrylhydarzyl (DPPH) free radical scavenging potential was higher (70.1%) in leaves extract than in callus extracts (46.3%) at the concentration of 10 mg/ml though both extracts had lower DPPH free radical scavenging activity compared to the positive control, vitamin C and BHT. Theresults conclude that the optimum concentration was IBA 1.5 mg/l+BAP 1.0 mg/l combination to produce callus cell proliferation and concentration of 2.0 mg/l IBA+0.25 mg/l BAP combination was the optimum for root proliferation of broccoli in vitro.

    The present study was conducted to evaluate the antioxidant activity of Vernonia cinerea, Peperomia
    pellucida and combination of Vernonia cinerea and Peperomia pellucida. These two herbs are pants that
    often grow at random in different environments but are not commercialized due to the fact that no
    comprehensive study of the importance of their use. The extract was prepared with methanol respectively.
    1,1-diphenyl-2-picrylhydrazyl (DPPH) assay were used to study their antioxidant activity. The extracts
    were compared with commercial antioxidant, butylated hydroxytoluene (BHT). The highest scavenging
    effect from peel extract was presented by Vernonia cinerea with the value of 76.3% scavenging activity
    (IC50 = 2.909), followed by the combination of Vernonia cinerea and Peperomia pellucida (71.21%
    scavenging activity; IC50 = 5.274) and Peperomia pellucida with value of 68.3% scavenging activity (IC50
    = 5.572). BHT showed the lowest IC50 value 1.71 with the scavenging activity 90.0%. Low IC50 value will
    indicates the strong ability of the extracts to act as DPPH scavenger.

    The present study was conducted to evaluate the antioxidant activity of peel extract from three types of
    melon, Cucumis melo var cantalupensis, Cucumis melo var inodorus and Citrullus lanatus in family
    Curcurbitaceae. The extract was prepared with methanol respectively. 1,1-diphenyl-2-picrylhydrazyl
    (DPPH) assay were used to study their antioxidant activity. The extracts were compared with commercial
    antioxidant, butylated hydroxytoluene (BHT). The highest scavenging effect from peel extract was
    presented by Cucumis melo var inodorus with the value of 52.7 ± 9.1µg/ml (IC50 = 4.61). BHT showed
    the lowest IC50 value 1.71 with the scavenging activity 90.0 ± 1.7µg/ml. Low IC50 value will indicates the
    strong ability of the extracts to act as DPPH scavenger.

    Introduction: Piper sarmentosum is one of the herbaceous plants that has been used as natural antioxidant to source to treat diseases. This study was conducted to determine the total phenolic contents (TPC) and free radical scavenging capacity in free and bound (soluble and insoluble) of P. sarmentosum. Methods: Free phenolic extract was acquired through direct methanol extraction while acidic and alkaline hydrolyses were adopted to release the bound phenolic acids. The TPC was determined by using Folin-Ciocalteu assay and is expressed as Gallic Acid equivalent (GAE) in miligrams per gram of extracts. The antioxidant scavenging capacity was determined by using DPPH (2, 2-diphenyl-1-picrylhydrazyl) assay. Results: Insoluble bound phenolic extract of P. sarmentosum showed the highest TPC value (1.54 ± 0.04 mg GAE/g DW) followed by soluble phenolic extract and free extract (1.13 ± 0.10 and 0.57 ± 0.06 mg GAE/g DW, respectively). The soluble phenolic fraction has expressed the highest free radical scavenging capacity (76.57± 4.12%) followed by insoluble (69.79± 2.33 %) and free extracts (58.15± 4.44 %). The IC50 values for free, soluble and insoluble bound phenolic were 24.05 ± 3.81, 16.17 ± 1.84 and 18.49 ± 1.92 mg/ml, respectively. Conclusions: The significant differences between all the extracts and antioxidant inhibition in this present study suggested that different forms (free and bound) of extracts did influence the radical scavenging capacity as a whole.

    Introduction: Clinacanthus nutans is used as natural nutraceuticals for prevention and treatment of cancer. The purpose of this study is to (i) determine the total phenolic content and antioxidant scavenging capacities of C. nutans in free and bound phenolic acid and (ii) study the relationship between TPC and antioxidant scavenging capacities of C. nutans. Methods: The total phenolic contents were measured using Folin-Ciocalteu assay. Free and bound phenolic were examined by using spectrophotometer while antioxidant capacity were evaluated using DPPH (2, 2-diphenyl-1-picrylhydrazyl) scavenging activity assay. Results: Insoluble phenolic acids showed the highest amount of total phenolic content in C. nutans extracts (6.09+ 0.45 mg gallic acid equivalent (GAE)/ g DW) and exhibited highest antioxidant activity (73.3+0.82 %) as compared to free and soluble phenolic extracts. The IC50 values for free phenolic, soluble bound and insoluble bound phenolic extracts were 0.69+0.02 mg/mL, 0.64+0.04 and 0.60+0.006 mg/mL, respectively. There were positive correlation between insoluble bound phenolic content of C. nutans extracts with antioxidant radical scavenging capacity (R2 = 0.893). Conclusions: These results indicate that different phenolic acid forms affect the total phenolic content and antioxidant properties. Natural compounds such as phenolics from C. nutans could be a good source of antioxidant.

    Balamuthia mandrillaris and Naegleria fowleri are free-living amoebae that can cause life-threatening infections involving the central nervous system. The high mortality rates of these infections demonstrate an urgent need for novel treatment options against the amoebae. Considering that indole and thiazole compounds possess wide range of antiparasitic properties, novel bisindole and thiazole derivatives were synthesized and evaluated against the amoebae. The antiamoebic properties of four synthetic compounds i.e., two new bisindoles (2-Bromo-4-(di (1H-indol-3-yl)methyl)phenol (denoted as A1) and 2-Bromo-4-(di (1H-indol-3-yl)methyl)-6-methoxyphenol (A2)) and two known thiazole (4-(3-Nitrophenyl)-2-(2-(pyridin-3-ylmethylene)hydrazinyl)thiazole (A3) and 4-(Biphenyl-4-yl)-2-(2-(1-(pyridin-4-yl)ethylidene)hydrazinyl)thiazole (A4)) were evaluated against B. mandrillaris and N. fowleri. The ability of silver nanoparticle (AgNPs) conjugation to enrich antiamoebic activities of the compounds was also investigated. The synthetic heterocyclic compounds demonstrated up to 53% and 69% antiamoebic activities against B. mandrillaris and N. fowleri respectively, while resulting in up to 57% and 68% amoebistatic activities, respectively. Antiamoebic activities of the compounds were enhanced by up to 71% and 51% against B. mandrillaris and N. fowleri respectively, after conjugation with AgNPs. These compounds exhibited potential antiamoebic effects against B. mandrillaris and N. fowleri and conjugation of synthetic heterocyclic compounds with AgNPs enhanced their activity against the amoebae.

    Garlic has been a favorite additive in food for many years in various cultures. It is known that garlic
    (Allium sativum) possesses antimicrobial, antiprotozoal, antimutagenic, antiplatelet and antihyperlipidemic
    properties. Allicin, a thiosulfinate extract of garlic, has been presumed to be a very strong antioxidant. High performance liquid chromatography (HPLC) analysis of raw garlic extract was not conclusive to determine allicin’s presence. However, using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging methods to determine the antioxidant activity of raw garlic extract shows a color change from deep violet to yellow, indicating antioxidant activity. Thus, raw garlic can be a source of antioxidant based on the results of the DPPH scavenging analysis.

    This study investigated the effects of different percentages of ethanol (0 - 100%), extraction times (1 - 5 h) and temperatures (25 - 60°C) on total phenolic content (TPC) and antioxidant activity (AA) of sapodilla pulp and peel. TPC was determined by Folin-Ciocalteu reagent method, while AA was evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay, 2,2-azino-bis-(3-ethyl-benzothiazoline-6-sulfonic acid) (ABTS) radical scavenging assay, ferric reducing antioxidant power (FRAP) assay and β-carotene bleaching (BCB) assay. Based on the optimal extraction conditions used, sapodilla pulp extract had TPC of 3.89 mg GAE/g, 63.20% of DPPH scavenging activity, 4.30% of ABTS scavenging activity, 19.17% of BCB activity, and FRAP value of 15.24 mg TE/g; while its peel extract had TPC of 9.23 mg GAE/g, 92.95% of DPPH scavenging activity, 5.36% of ABTS scavenging activity, 8.14% of BCB activity, and 27.85 mg TE/g (FRAP value). Using the optimal extraction conditions for sapodilla pulp (40% ethanol as extraction solvent that extracted at 60°C for 4 h) and sapodilla peel (80% ethanol and 2 h extraction time at 40°C), highest antioxidants can be extracted from the pulp and peel.

    Antioxidants in seaweeds have attracted increasing interest for its role in protecting human health. Therefore, the aim of this study was to assess the Total phenolic content (TPC) values and antioxidant activities in red seaweeds Kappaphycus alvarezii and Kappaphycus striatum of different solvent extracts. Total phenolic content (TPC) and antioxidant activities (DPPH scavenging assay and Trolox equivalent antioxidant capacity assay, TEAC) for both K. alvarezii and K. striatum extracts were determined using different solvents at different concentrations (ethanol: 50%, 70%, 100%; acetone: 50%, 70%, 100%; methanol: 50%, 70%, 100%). The TPC value was measured using the Folin-Ciocalteu’s method. The antioxidant activities were measured by 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay and Trolox Equivalent Antioxidant Capacity (TEAC) assay. The highest TPC value of K. alvarezii antioxidant extract was obtained by 50% ethanol extracts while for K. striatum obtained by 50% methanol extract. The highest percentage of DPPH free radical inhibition for K. alvarezii was shown by 50% acetone extract while K. striatum was shown using 50% methanol extract. The highest TEAC value for K. alvarezii was shown by 50% acetone while K. striatum extract was shown by 50% ethanol extract. The TPC values and antioxidant activities of all solvent extracts of K. striatum were significantly higher (p< 0.05) than K. alvarezii antioxidant extracts. The TPC values showed strong correlation (r = 0.797) with TEAC values for K. alvarezii antioxidant extract (p< 0.01). The TEAC values also showed strong correlation (r = 0.735) with percentage of DPPH free radical inhibition for K. alvarezii (p< 0.01). The TPC value, DPPH free radical scavenging assay and TEAC assay for K. striatum extracts showed strong correlation (r> 0.8) with each other (p< 0.01). In summary, K. striatum showed better antioxidant activity and higher TPC value than K. alvarezii.

    This study was done to assess the total phenolic compounds (TPC) and 1, 1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity in the flowers and leaves of Clitoria ternateaand Vitex negundo Linn. by using methanol and water extraction. TPC were evaluated using Folin-Ciocalteau method. Methanol was more efficient in extracting phenolic compounds compared with water in measuring TPC. Vitex negundo Linn. contained higher amount of TPC compared to Clitoria ternatea. Besides that, leaves for both plants showed higher amount of TPC compared to the flowers. Methanol extracted Vitex negundo Linn. showed higher DPPH scavenging activity compared with Clitoria ternatea. In contrast, DPPH scavenging activity for water extracted Clitoria ternatea showed higher value in compare with water extracted Vitex negundo Linn. The type of solvent used to extract the plant material and concentration of extracts used showed significance difference (P < 0.05) on the amount of DPPH scavenged by the plant extract. The presence of antioxidant activity in both leaves and flowers showed that Clitoria ternatea and Vitex negundo Linn. have the potential to be an alternative source of natural antioxidants. In vivo study is needed for successful commercialization and to benefit both the food and pharmaceutical industries.

    The effects of ethanol concentration (0-100%, v/v), extraction time (60-300 min) and extraction temperature (25-65°C) on the extraction of phenolic antioxidants from Andrographis paniculata was evaluated using single-factor experiments. The following complementary assays were used to screen the antioxidant properties of the crude extracts: total phenolic content (TPC), total flavonoid content (TFC), condensed tannin content (CTC), 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical-scavenging capacity and 2,2’-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging capacity. The extraction conditions chosen had significant effects (p < 0.05) on the extraction of phenolic compounds and antioxidant capacity. The optimal conditions were 60% ethanol for 60 min at 65oC for phenolic compounds and at 25oC for antioxidant capacity. Strong negative significant (p < 0.05) correlations were observed between the phenolic compounds (TPC, TFC and CTC) and antioxidant capacity comprising ABTS (-0.924, -0.909, -0.887, respectively) and DPPH radical-scavenging capacities (-0.992, -0.938, -0.928, respectively) were determined under the influence of extraction temperature.

    Papaya (Carica papaya L. cv. Hongkong) is an economically important fruit crop grown in Malaysia. During its ripening stages, (C. papaya L.) exhibits different physicochemical properties, antioxidant capacities, and sensory quality results. The objective of this study was to elucidate in detail the antioxidant capacity of C. papaya as determined by total phenol content (TPC), total flavonoid content (TFC), ferric reducing antioxidant power (FRAP),2,2-diphenyl-1-picrylhydrazyl (DPPH) and scavenging systemand (ABTS). The study also aimed to study physicochemical changes of papaya fruits based on measured pH, titratable acidity (TA), total soluble solids (TSS), moisture and fruit color at five different stages of ripening. The fruits were harvested at five different, stages RS1, RS2, RS3, RS4, and RS5 corresponding to 12, 14, 16, 18, and 20 weeks after anthesis, respectively. Significant differences were found at different stages of ripening. The pH of the fruit decreased significantly (P < 0.05), whereas TA, moisture, and TSS increased significantly (P < 0.05) during the ripening process. The redness (a*) and yellowness (b*) values of fruit color both increased significantly (P < 0.05), whereas
    lightness (L*) varied. The total phenol content TPC, TFC, FRAP, DPPH and ABTS values increased significantly (P < 0.05) with the ripening process. Sensory evaluation based on the color, sweetness, sourness, flavor, and overall acceptance for the last three maturity stages was also performed. RS5 had a better score than RS3 or RS4. The results showed the important role of the ripening stage in increasing the antioxidant content of papaya fruits.

    Antioxidant capacity of Ocimum tenuiflorum L. or ‘ruku’ were determined in this study. Fresh
    leaves of Ocimum tenuiflorum was subjected to freeze drying, vacuum drying and processed
    into fermented and unfermented tea. The samples were extracted using distilled water and the
    total phenolics, total flavonoids, condensed tannin content, anthocyanins and total antioxidant
    capacity (TAC) were assessed, measured with ferric reducing antioxidant power (FRAP)
    and 1,1-diphenyl-1-picrylhydrazyl radical scavenging capacity (DPPH) assays. The results
    showed that drying the fresh leaves of Ocimum tenuiflorum and processing them into tea leaves
    significantly increase (P < 0.05) the antioxidant capacity, total phenolic content, total flavonoid
    content, and condensed tannin content. However, anthocyanins content showed reduction
    after drying. In the present study, it can be concluded that the vacuum drying method seem
    to produce a product with higher quality of antioxidant properties than freeze drying. Hence,
    vacuum drying can be used to replace freeze drying as it is also cheaper than freeze drying.

    The effects of squid ink at concentration of 0.10 and 0.25% on the total bacteria count and
    chemical spoilage indicator; total volatile basis nitrogen (TVBN) and trimethylamine (TMA)
    of squid (Loligo duvauceli) were analysed. The analysis were performed at interval of 5 days
    during 15 days of chilled storage (4°C). This studies also investigate the antioxidant capacity
    of the squid ink. The melanin-free squid ink were subjected to ferric reducing power (FRAP)

    The effects of sodium chloride (NaCl) (3.5%) solution and polysaccharides, such as carboxymethyl cellulose (CMC) (0.1, 0.3 and 0.5%) and gum arabic (5, 10 and 15%), on the physicochemical properties, antioxidant capacity and sensory characteristics of bitter gourd juice were investigated. An increase in the concentration of CMC and gum arabic significantly was observed to increase the lightness (L value) and the viscosity (mPas) of bitter gourd juice at all levels. Increased concentrations of gum arabic significantly increased the total soluble solids. The bitter gourd fruit treated with NaCl solution produced the highest lightness (L value) and scavenging activity of free radical 2,2-diphenyl-1-picrylhydrazyl of bitter gourd juice. Increased concentration of gum arabic up to 15% significantly increased the total phenolic content. The addition of 5% gum arabic effectively reduced the bitterness of the bitter gourd juice. Viscosity of the juice resulted in negative correlation for bitterness.

    Keupayaan Centella asiatica (CA) untuk bertindak sebagai antioksidan dan agen anti-radang telah banyak dilaporkan. Namun begitu, kaedah pengekstrakan CA untuk memperoleh hasil yang terbaik masih dipersoalkan. Dalam kajian ini, kami menilai tiga kaedah pengekstrakan CA dan membuat perbandingan ekstrak dari segi aktiviti antioksidan dan anti-radang, dan juga kandungan sebatian bioaktif, asiaticoside dan madecassoside. Centella asiatica diekstrak menggunakan pelarut etanol, metanol dan juga air. Kandungan sebatian fenolik ekstrak diukur menggunakan kaedah reagen Folin-Ciocalteu. Kandungan asiaticoside dan madecassoside ditentukan dengan kaedah HPLC. Aktiviti antioksidan diukur dengan asai 2,2-diphenyl-1-picrylhydrazyl (DPPH) dan asai penurunan kuasa Ferric Reducing Antioxidant Power (FRAP). Aktiviti anti-radang ditentukan dengan kebolehan ekstrak untuk merencatkan enzim tapakjalan keradangan, COX-1 dan COX-2, serta kebolehan ekstrak melindungi sel fibroblas aruhan 12-O-tetradecanoylphorbol-13-acetate (TPA) daripada menghasilkan prostaglandin E2 (PGE2 ). Hasil kajian menunjukkan aras sebatian fenolik, asiaticoside dan madecassoside tertinggi dalam ekstrak etanol, diikuti metanol dan esktrak akues (masing-masing 17.76 g/100g, 15.52 g/100g, 13.16 g/100g untuk sebatian fenolik, 42.86 mg/g, 36.37 mg/g, 2.82 mg/g untuk asiaticoside and 18.66 mg/g, 15.87 mg/g, 3.75 mg/g untuk madecassoside). Ketiga-tiga ekstrak menunjukkan aktiviti antioksidan sederhana berbanding kawalan positif. Kesemua ekstrak, asiaticoside dan madecassoside merencat COX-1 dan COX-2 dan menyekat penghasilan PGE2 -aruhan TPA. Ekstrak etanol dan metanol merupakan perencat COX yang lebih kuat dan lebih poten daripada ekstrak akues. Oleh itu, walaupun ekstrak akues menunjukkan kebolehan antioksidan yang lebih tinggi, dari segi aktiviti anti-radang, pelarut hidrofobik iaitu etanol dan metanol ternyata lebih baik untuk mengekstrak Centella asiatica.

    The extraction of phenolics from Citrus hystrix leaf was carried out using supercritical fluid extraction and was optimized using response surface methodology (RSM). The effects of CO2 flow rate, extraction pressure and extraction temperature on yield, total phenolic content and diphenyl-picrylhydrazyl-IC50 were evaluated and compared with ethanol extraction. The extraction pressure was the most significant factor affecting the yield, TPC and DPPH-IC50 of the extracts, followed by CO2 flow rate and the extraction temperature. The optimum conditions of pressure, CO2 flow rate and temperature were at 267 bars, 18 g/min and 50°C, respectively.     Total phenolic content (TPC) and antioxidant properties of xanthone extract from mangosteen pericarp via microwave-assisted extraction (MAE) method was optimized by response surface methodology (RSM). The MAE extraction conditions to obtain optimum antioxidant-rich xanthone extract were at 2.24 min of irradiation time, 25 mL/g of solvent-to-solid ratio and 71% of ethanol concentration. The predicted results for four responses were as follows; 320.31 mg gallic acid equivalent/g extract, 83.63% and 93.77% inhibition (DPPH (2,2-diphenyl-1-picrylhydrazyl) and ABTS (2,2'-Azino-bis-3-ethylbenzthiazoline-6-sulfonic acid) assays), and 144.56 mg Trolox equivalent/g extract (FRAP, Ferric reducing antioxidant power). The predicted and actual values were statistically insignificant (P > 0.05). Therefore, these results confirmed that the examined model was acceptable and relevant. MAE led to a slightly similar antioxidant capacity and a higher extraction of α-mangostin, a major xanthone of mangosteen pericarp as compared to water bath-maceration technique.
